Tissue Culture Laboratory

Plant tissue culture plants must be incubated in a sterile chamber after the initiation or inoculation process. In incubation room, the plants can grow well if the light, temperature, and humidity are properly adjusted. Some plant tissues grow well under low light condition (approx. 1000 lux). Light and dark periods are needed for regeneration. Regenerated plant need well lighted (approx. 3000 lux) condition with 16 hours light and 8 hour dark period. The illumination in the culture room is provided by cool white fluorescent light placed approximately 18″ above the culture racks. The temperature are set on the range between 20-25°C. The humidity are set on the range of 50-80%. The sterile condition of this room is important to prevent contamination, especially from dust or insect. Therefore, alcohol 70% must be sprayed regularly.
